Research Expertise
Description
My research programme is focused on obtaining an increased understanding of the physicochemical properties of active and excipient materials used in a variety of different dosage forms, and on using that knowledge to improve product design in terms of in vitro and subsequently in vivo performance. This research objective is achieved through collaborative efforts and interdisciplinary research with both academic and industry partners, nationally and abroad. My research field is aligned with "Therapeutics: Synthesis, Formulation, Processing and Drug Delivery" which is one of 14 priority areas for research identified under the National Research Prioritisation Exercise (NRPE). My current research spans three principal strands: (i) Pharmaceutical material science (ii) Particle engineering for pulmonary drug delivery, and (iii) Enabling formulations for poorly soluble and poorly bioavailable therapeutic agents (i) Pharmaceutical material science Research under this theme is aimed at understanding how solid state and micromeritic characteristics of drug and excipient materials are impacted by pharmaceutical processing, and how formulation and/or processing may be optimized so as to control physicochemical characteristics and drug product performance for oral solid dosage forms. My expertise in this area has led to me taking on the role of Co-PI and Strand Leader in the Synthesis and Solid State Pharmaceutical Centre (SSPC), an SFI-industry co-funded research centre. (ii) Particle engineering for pulmonary drug delivery Sophisticated particle engineering processes enable improved delivery of powders via oral inhalation. In my group, we have developed an internationally recognised expertise in formulation and particle engineering by spray drying for dry powder inhalation. This expertise led to me being invited to join an international collaborative translational NIH-funded 8 million project, led by the University of California San Francisco, aimed at developing a novel mucolytic therapy for the treatment of lung disease, including cystic fibrosis and asthma. The project commenced in 2016. (iii) Enabling formulations for poorly soluble and poorly bioavailable therapeutic agents It is estimated that more than 60% of new drug molecules display poor aqueous solubility, due to an increased size and lipophilicity. There is a need for enabling formulations for such therapeutic agents which have suboptimal bioavailability. Alterations of the solid state form by means of salification, cocrystallization or amorphisation are options which we have explored, as is microencapsulation for controlled release. My group's expertise in this area forms the basis for a number of projects in the SSPC and in AMBER (Advanced Materials and BioEngineering Research centre), another SFI-industry funded research centre, where I am a funded investigator.Recognition
